The Acts are all structured so that many of the quests have to be done in a specific order anyway. From personal experience, it took me around ~30 hours to finish the story of DAO alone, I reckon with Awakening that's 50 overall + other dlcs? DA II was roughly the same for the base game since I never owned the DLCs. Buuuuut, if my reinvigorated Dragon Age high remains strong, I might actually do a trilogy run.

At the end of act 2 is supposed to be the best time to play Mark of the Assassin, then act 3 is helpful in act 3 just before you wind up getting into the end cause it adds bonus to the end fight against Meredith and the templars.
The DLC gear that you get from the chests in Gamlen's place looks pretty strong, and some of them scale with you as you level up. Can that gear take me to end game? As well as Anders, as stated by whyihatepink below. He is affected by the end boss due to being a Grey Warden and hearing the call of the darkspawn below. An alt for Anders would be Varric as there is a pretty neat side quest for him there. However, if I remember correctly each character has a side quest. Nope, thats MotA.. sorry.
